How to Use the Yeast Plasmid Rapid Extraction Kit

Yeast Plasmid Extraction:

Add 20 µL of One-Step Yeast Plasmid Extraction Buffer to a PCR tube.

Use a sterile pipette tip or toothpick to pick a small amount of yeast colony and add it to the PCR tube.

Vortex or gently mix to combine.

Heat at 98°C for 20 minutes.

Spin down at 4,000 rpm for 5 minutes to collect the supernatant.

Transfer the supernatant to a clean PCR tube without disturbing the pellet.

Transformation and Identification (Optional, if using for transformation):

Thaw competent cells on ice.

Add 5 µL of the extracted yeast plasmid to 100 µL of competent cells, mix gently (avoid vigorous shaking).

Incubate on ice for 30 minutes.

Heat shock at 42°C for 1 minute, then place back on ice for 2 minutes.

Add 900 µL of SOC medium (no antibiotics) and shake at 37°C for 1 hour.

Spin at 4,000 rpm for 5 minutes, discard 800 µL of supernatant, resuspend the pellet, and plate onto selective agar plates.

Incubate at 37°C overnight.
