Human NT5E Knockout A-431 Cell Line

DESCRIPTION

Human NT5E Knockout A-431 Cell Line is engineered from human epidermoid carcinoma A-431 cells using CRISPR/Cas9-mediated disruption of the 5′-Nucleotidase Ecto (NT5E/CD73) gene, a cell surface enzyme that converts extracellular AMP to adenosine, playing pivotal roles in purinergic signaling and immune suppression. This knockout model exhibits abolished adenosine production and impaired immunosuppressive activity while maintaining A-431 cells’ characteristic epithelial morphology. The cell line serves as a valuable tool for studying cancer immunotherapy resistance, tumor microenvironment remodeling, and inflammatory responses, particularly in research involving A2A adenosine receptor signaling and checkpoint inhibitor efficacy. Maintained at low passage numbers (<P20) with genetic stability, the knockout efficiency is validated through genomic PCR, Sanger sequencing, and functional assays demonstrating loss of AMP hydrolysis activity. Rigorous quality control confirms the cell line is free of contamination from HIV-1, HBV, HCV, Syphilis, mycoplasma, fungi, yeast, and bacteria.
