Principle of the Assay: NEFA ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-
NEFA antibody and an NEFA-HRP conjugate. The assay sample and buffer are incubated together with NEFA-HRP conjugate
in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times.
The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction
forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow.
The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is
inversely proportional to the NEFA concentration since NEFA from samples and NEFA-HRP conjugate compete for the
anti-NEFA antibody binding site. Since the number of sites is limited, as more sites are occupied by NEFA from the sample,
fewer sites are left to bind NEFA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.)
to the concentration of standards. The NEFA concentration in each sample is interpolated from this standard curve.
Intended Uses: This NEFA ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine NEFA.

Catalyzes the removal of terminal sialic acid residues from viral and cellular glycoconjugates. Cleaves off the terminal
sialic acids on the glycosylated HA during virus budding to facilitate virus release. Additionally helps virus spread
through the circulation by further removing sialic acids from the cell surface. These cleavages prevent self-aggregation
and ensure the efficient spread of the progeny virus from cell to cell. Otherwise, infection would be limited to one
round of replication. Described as a receptor-destroying enzyme because it cleaves a terminal sialic acid from the
cellular receptors. May facilitate viral invasion of the upper airways by cleaving the sialic acid moities on the mucin
of the airway epithelial cells. Likely to plays a role in the budding process through its association with lipid rafts
during intracellular transport. May additionally display a raft-association independent effect on budding.
Plays a role in the determination of host range restriction on replication and virulence. Sialidase activity in
late endosome/lysosome traffic seems to enhance virus replication.
